The Quansys multiplex ELISA, known as the Q-Plex technology, is a fully quantitative ELISA-based test where up to 25 distinct capture antibodies have been deposited to each well of a 96 or 384-well plate in a defined array. Each spot represents a distinct capture antibody population.
Spotting Antibody in 96 Well Plate
- Robotic liquid handlers print 20-50nl spots of capture antibody
- Each spot is a unique assay within the well
- Spot to spot variability<4% CV
- Spot size 350-500 µm
Performing the Assay
- Add 30 µl of sample
- Wash
- Add mix of detection antibodies
- Wash
- Add streptavidin bound to a IR-Dye or HRP
Detection of Sample
- Each spot is a different assay (IL-1, IL-2, etc.)
- With the addition of substrate, a response is produced
- If antigen is present the spot emits light
- If no antigen is present the spot is not visible
Image Capture
- An image is taken of the plate via high resolution camera or fluorescent scanner
- The image file (TIFF) is imported into Q-View Software
Image Analysis
- Image is opened in Q-View Software
- Spots are automatically found on plate image
- Intensity of spot response is measured and raw data is generated
- User imports product specs and well layout
Data Analysis
- Raw data is analyzed and compared to in-plate standard
- Regression models used to calculate unknowns
- Standard curves are calculated and sample and statistical data is exported

Using less than 30 µl of sample, up to 84 different samples can be assayed for all 25 unique analytes in less than 2.5 hours using one ELISA test kit. Sensitivity is system dependent, typically ranging between 30 pg/ml to less than 1 pg/ml. All of the antibodies used in the Q-Plex Arrays have been subjected to a rigorous and comprehensive cross reactivity protocol and verified to be non-cross reactive with any other system in the array. Detection of this array is performed using the Q-View Imager or other alternative imaging systems. The image is then auto-processed using Q-View Software and sample concentrations for each analyte are reported.
