coronavirus Research Tools

Quansys Biosciences has released two new products to help aid in the research of SARS-CoV-2, the novel coronavirus causing COVID-19. The first is a human serological 4-plex assay that simultaneously detects IgG class antibodies to both the S1 and S2 subunit of the SARS-CoV-2 spike protein, with a positive and negative control in each well. The second is a Cytokine Storm panel that quantitatively measures 16 different cytokines related to Cytokine Release Syndrome associated with COVID-19 infections, including IL-6, Interferon alpha and Interferon gamma. These products provide researchers an easy-to-use and cost-effective means of generating a cytokine profile for each sample and detecting the presence of antibodies in each sample.

SARS-CoV-2

The SARS-CoV-2 Human IgG (4-plex) assay is a qualitative ELISA that detects IgG antibodies to the SARS-CoV-2 virus in human serum or plasma. The virus has several structural proteins, including two spike proteins, S1 and S2. When an individual is infected with the SARS-CoV-2 virus, their immune system produces antibodies to these viral proteins. The typical immune response produces detectable antibody levels ~8-10 days following the onset of symptoms. Our assay detects IgG antibodies to both the S1 and S2 spike proteins present in a blood sample. As a result of testing for both proteins, this is one of the best performing assays on the market based on FDA calculation tools, with results of 100% PPA and 99.7% PPV with a 5% prevalence used in the calculation. If antibodies are detected, the result will be considered positive. If antibodies are not detected, the result will be considered negative.

Cytokine Release Syndrome

Cytokine Release Syndrome (CRS) is acute systemic inflammation associated with COVID-19 infections, as well as treatments from therapeutic antibodies, CAR-T cell therapy, and even haploidentical marrow transplantations.  During CRS, a large number of cytokines are rapidly released into the bloodstream from immune cells affected by the virus.  Symptoms may range from headache and fever to organ damage or even death.  The Q-Plex™ Human Cytokine Release Syndrome (16-plex) kit is a quantitative ELISA-based chemiluminescent assay designed for the concurrent measurement of 16 distinct analytes in the same well from only 25 uL of sample.  In under five hours, up to 80 samples can be tested on the same microplate.

SARS-CoV-2 Human IgG (4-Plex)

The SARS-CoV-2 Human IgG (4-Plex) multiplex assay allows for simultaneous indirect ELISA on the following four printed spots:

1- SARS-CoV-2 Spike Glycoprotein (S1), a recombinant antigen which contains amino acids 1-674 of subunit 1. Spike S1 is expressed in mammalian HEK293 cells with a Sheep Fc-Tag.

2- SARS-CoV-2 Spike Glycoprotein (S2) is a recombinant antigen which contains the Spike subunit 2 protein, amino acids 685-1211. Spike S2 is expressed in mammalian HEK293 cells with a Sheep Fc-Tag.

3- Sheep Fc, is a negative control to ensure no cross-reactivity occurs between human IgGs in the sample and the Fc-Tag on the SARS-Cov-2 Spike proteins.

4- Anti-Human IgG is a positive control to ensure the kit performs and the IFU was followed correctly.
