Human P-Selectin

P-selectin, also known as CD62P, Granule Membrane Protein 140 (GMP-140), and Platelet Activation-Dependent Granule to External Membrane Protein (PADGEM), is a protein that plays an important role in immune responses and inflammation. Unactivated epithelial cells that line blood vessels hold P-selectin in granules called Weibel-Palade bodies and unactivated platelets store it in α-granules. When activated by histamine or thrombin during an immune response, the granules release P-selectin to the surface of the cell, where it acts as a cell adhesion molecule (CAM) that works to recruit white blood cells and other platelets. The protein promotes aggregation in platelets and is critical for stopping bleeding at the site of an injury. P-selectin also has a critical role in cancer, particularly tumor metastasis. During metastasis, P-selectin helps migrating cancer cells enter the bloodstream and form a complex with platelets that makes it difficult for macrophages to recognize the cancer. Inhibition of P-selectin is a target of therapeutic research to prevent tumor metastasis.
