Matrix metallopeptidase 9 (MMP9), also known as type IV collagenase, gelatinase or gelatinase B (GELB), is a member of the MMP family of enzymes involved in degrading the extracellular matrix. MMP9 is involved in many processes, including early development, blood vessel growth, wound healing, and cell migration. In immune responses, MMP9 enables neutrophil migration along the basement membrane by breaking down the cellular matrix and activating critical cytokines. During development and wound healing, it plays an important role in angiogenesis and barrier formation, recruiting endothelial cells to form blood vessels and epithelial cells to close wounds. As with many members of the MMP family, dysregulation of MMP9 is associated with numerous pathologies. Elevated levels are associated with risk for arthritis, idiopathic atrial fibrillation, aortic aneurisms, and many forms of cancer where the enzyme enables tumor formation and growth. Genetic variants of MMP9 have been identified that help prevent the transfer of malaria from mother to fetus, suggesting it may play a role in susceptibility to the parasite.
